Evaluación de antígeno leptospiral con suero hiperinmune humano y animal mediante microELISA y microalutinación. Antigen of leptospirosis, its assessment with human and animal hyperimmune serum through MicroELISA and microagglutinationEl ELISA ha demostrado alta sensibilidad para el diagnóstico de diversas enfermedades y patologías y también para el diagnóstico serológico de leptospirosis. Muchos investigadores han elaborado antígenos de distintas cepas leptospirales con propósitos diagnósticos, con resultados satisfactorios. Este trabajo tiene como objetivo evaluar un antígeno leptospiral elaborado con una cepa virulenta utilizando la técnica de microELISA comparándola con la microaglutinación que es la técnica de referencia de la OMS. Se estudiaron cuatro sueros hiperinmunes de pacientes con leptospirosis de los cuales se aislaron cuatro cepas, cuatro sueros de cordón umbical de la sección SUMA Hospital «Pepe Portilla», nueve sueros hiperinmunes de consejos y diecinueve sueros de trabajadores de la Facultad de Ciencias Médicas antes de vacunarse. Para la microaglutación se emplearon veinte serogrupos de leptospiras enfrentándoles a los sueros diluidos desde 1:50 hasta 1:25600 y para microELISA se diluyó desde 1:2 hasta 1:256. E n microELISA se empleó un policlonal de IgG, IgA e IgM marcado con peroxidasa de la firma SIGMA suministrado por el laboratorio de leptospirosis del IPK. Los resultados coincidentes 100% entre ambas técnicas respecto a los sueros hiperinmunes humanos, 100 % no coincidentes con los conejos, 100% con los sueros controles negativos de cordón umbical y 94.8 % coincidentes en los sueros de trabajadores tanto positivos como negativos. Se concluye que el antígeno utilizado se fijó excelentemente a las placas para microELISA. Se demostró la heterología de la región FC de las IgG de conejo y las inmunoglobulina anti FC humanas. También la coincidencia de casi 95% entre ambas técnicas para el diagnóstico normal brinda ventajas técnicas, económicas y sociales. DeCs: MicroELISA, LEPTOSPIROSIS, INVESTIGACION ABSTRACT The ELISA has proved higly sensitive to the diagnosis of several diseases and pathologies, as well as to the serological diagnosis of leptospirosis. Many researchers have made antigens from different leptospiral strains for diagnostic purposes obtaining satisfactory results. The objective of this paper is to assess the leptospiral antigen made from an avirulent strain by using a microELISA technique of WHO. Four hiperimmune sera from patients with leptospirosis were studied.Four strains, four umbilical cord sera from SUMA department of Pepe Portilla Hospital, nine hiperimmune sera from rabbit and 19 sera from workers of the Faculty of Medical Sciencies (before vaccination) were isolated. Twenty leptospiral serogroups against the diluted sera from 1:50 to 1:25600 were used for microagglutination, whereas microELISA was diluted from 1;2 to 1:256. For microElisa, we used an IgG, IgA and IgM policlonal marked with peroxidase (SIGMA) by IPK leptospirosis laboratory. Identical results (100%) were obtained in both techniques with respect to human hiperimmune sera, 100% was not consistent with those of the rabbits, 100% with the sera of umbilical cord from negative controls and 94.8% were consistent with sera from workers (disregarding their positiveness or negativeness). It is concluded that the antigen used had an excellent fixation to the slides for microELISA. Heterology of FC region from rabbit's IgG and immune globulines anti FC humans was shown. Technical, economic and social advantages can be achieved taking into account the coincidence (95%) between both techniques for normal diagnosis.
