Identification of Cytochrmoe oxidase p450 in Heptocytes generated from in vitro differentiation of mouse mesenchymal stem cells

SUMMARY

This study was designed to identify the presence of cytochrome oxidase p450 (CYP3A4), in the hepatic cell lines which was generated from the in vitro differentiation of mouse bone marrow mesenchymal stem cells. Bone marrow meenchymal stem cells were first isolated by collecting both the femur and tibia of the mouse thigh bone, then exess tissue were removed and the cells were flushed from the bones and cultured under highly aseptic conditions. The isolated cells were characterized as a mesenchymal stem cells via using immunocytochemistry analysis. The characterized hepatocytes were subjected to a differentiation protocol in which the differentiation medium was contained a spesfic growth factor including (Hepatocytes growth factor (HGF), Fibroblast growth factor4 (FGF4), Oncostatin M and Dexamethasone, All theses growth factor was used for directing the BM-MSCs twards the hepatic line of cells. After three weeks of the differentiation protocol the diffrentation cells were examined for the presence of Cytochrome oxidase p450 (CYP3A4), which consider as one of the specific markers of hepatocytes. Identification eas done via two different methods first, via using the immunocytochemical analysis for the antihuman mouse CYP3A4 antibody and second by ELIZA test. The results showed a posotive reactivity of the differentiated cells towards the mouse antihuman CYP3A4 antibody as well as the eliza results showed a significant increase in the enzyme level of the cell lysate during the weeks of the differentiation experiment.

Tumor adenomatoide de epidídimo. Presentación de caso Tumor adenomatoide de epidídimo. Presentación de casoIntroducción: El tumor adenomatoide es el más común de los tumores sólidos del epidídimo. Es de naturaleza benigna y aún no se conoce con certeza su origen. Objetivos: Presentar un nuevo caso de tumor adenomatoide del epidídimo, y la secuencia diagnóstica y terapéutica empleada. Presentación del caso: Paciente de 39años de edadcon antecedentes de salud, que asistió a la consulta externa de Urología de la Clínica Multiperfil de Luanda, por aumento de volumen indoloro del hemiescroto derecho de 4 años de evolución. No refirió traumatismos ni procesos inflamatorios genitales previos. Al examen físico se confirmó la presencia de un tumor esférico, en la cabeza del epidídimo derecho, de unos 4 cm de diámetro de consistencia firme. El testículo homolateral, así como el epidídimo y el testículo contralaterales, eran normales. La ecografía reveló una masa heterogénea, bien delimitada y poco vascularizada. La resonancia magnética nuclear denotó una lesión hipointensa en imágenes ponderadas en T 1, e hiperintensa en imágenes ponderadas en T 2, tras la administración del contraste.  La biopsia por aspiración con aguja fina fue negativa de células neoplásicas. Se realizó la exéresis total de la lesión, vía transescrotal, sin complicaciones. El informe histopatológico de la pieza quirúrgica corroboró tumor adenomatoide. El paciente evolucionó satisfactoriamente. Conclusiones: El tumor adenomatoide de epidídimo es infrecuente. La ecografía y la resonancia magnética nuclear son útiles para el estudio imagenológico. La biopsia por aspiración con aguja fina permite obtener el diagnóstico histológico preoperatorio. It is of benign nature and is not well known its origin yet. Objective: to show a new case of adenomatous tumor of the epididymis, the diagnostic sequence, used therapeutic procedure and the differential diagnostic. Case presentation: patient of 39 years old with antecedents of health that assisted to the outpatient clinic of Urology in the Multi proposal Clinic of Luanda, because increased painless and volume of the right hemi scrotal sac of 4 years of evolution. It did not refer traumatisms or previous inflammatory genital processes. To the physical examination confirmed the presence of a spherical tumor in the head of the right epididymis, around 4 cm of diameter with firm consistency. The homolateral testicle, as well as the contralateral epididymis and the testicle, were normal. The echography revealed a heterogeneous mass, well delimited and little vascularized. The nuclear magnetic resonance denoted an tumor low intense in weighted images in T1, and high intense in weighted images in T2 after the administration of the contrast. The fine needle aspirate biopsy was negative of neoplastic cells. The total excision of the tumor was performed, road Trans scrotal, without complications. Histopathological report of the surgical piece corroborated adenomatous tumor, the patient going on satisfactorily. Conclusions: the adenomatous tumor of epididymis is infrequent. The echography and the Magnetic Resonance Image are useful for the image study. The fine needle aspirate biopsy allows to obtain the pre-surgical histological diagnostic. Eficacia del implante de células mononucleares autólogas en el tratamiento de la periodontitis crónica Eficacia del implante de células mononucleares autólogas en el tratamiento de la periodontitis crónicaIntroducción: Los buenos resultados en la regeneración de tejidos mediante la implantación de células mononucleares  en distintas especialidades médicas en el mundo y en Cuba, ha motivado a considerar el periodonto como candidato para esta terapia. Objetivo: Determinar la eficacia del implante de células mononucleares autólogas en el tratamiento de la periodontitis crónica a 48 meses de evolución. Material y Métodos: Se realizó un ensayo clínico fase III, aleatorizado y controlado en pacientes con periodontitis crónica durante los años 2008 a 2014. De ese grupo de pacientes se evaluaron 30 que tenían 48 ó más meses de recibir tratamiento en la consulta de Periodoncia del Hospital  “Enrique Cabrera” en el período comprendido de febrero a mayo de 2015; 15 habían recibido tratamiento con células mononucleares (grupo estudio) y 15 terapia convencional (grupo control). La movilización a la sangre periférica de las células mononucleares se realizó mediante la aplicación del factor estimulador de colonias de granulocitos (FEC-G). Resultados: Se aprecia una disminución del promedio de todas las variables clínicas, que es marcada en cada uno de los períodos evolutivos con respecto al estado inicial de los pacientes a los 6, 12,  36 y 48 meses posteriores al tratamiento en el grupo de estudio, mientras que en el control las manifestaciones de la enfermedad fueron reapareciendo paulatinamente a medida que avanzaba el tiempo post-tratamiento. Estos resultados fueron significativos. Conclusiones: La terapia celular regenerativa con células mononucleares autólogas es eficaz para el tratamiento de pacientes con periodontitis y su efecto es prolongado. Palabras clave: Periodontitis, terapia celular, células madre, regeneración periodontal, células mononucleares autólogas. ABSTRACTIntroduction: Over the last years, in Cuba and other countries, encouraging results have been obtained in different medical specialties in the field of tissue regeneration by implanting stem cells; it has motivated us to consider periodontal tissue as a target for this therapy. Objective: To determine the efficacy of the implant of mononuclear autologous cells for the treatment of 48 months evolution chronic periodontitis. Material and Methods: 30 patients attending the Periodontology Clinic of the “Enrique Cabrera” hospital for over48 months during years 2008 to 2014 where included in the study, divided into two groups 15 patients each: group A were treated with autologous mononuclear cells and group B, who underwent classic therapy. The peripheral blood autologous mononuclear cells (MNC-SP) were mobilized with granulocyte colony- stimulating factor (G-CSF). Results: Group A patients showed a significant average decrease in all clinical variables, more remarkable in every of the evolution periods as compared to the initial stage of every patient at 6, 12, 24 and 48 months after starting the treatment. In Group B patients, signs of the disease gradually reappeared as the post-treatment period proceeded. Conclusions: Regenerative cell therapy with MNC-SP is effective and long lasting in patients with periodontitis.Keywords: Periodontitis, cell therapy, stem cells, periodontal regeneration, bone marrow mononuclear cells.
