Potensi Ekstrak Solanum betaceum sebagai Agen Protektif Sel Leydig pada Mencit yang Dipapar Timbal Asetat

SUMMARY

Lead exposure as environmental pollution can cause male reproductive problems. Provision of natural antioxidants can be an alternative solution to protect male reproductive function. This research was conducted to investigate the potential of Solanum betaceum extract as a protective agent for Leydig cells in mice exposed to lead acetate. The entire research process with a replication unit of 40 male mice. Mice were divided into five groups with each treatment given orally. The negative control group (K0) only gets aquadest for 35 days, the positive control group (K1) gets aquadest day 1-3, day 4-35 get lead acetate at a dose of 0.075 mg/kg BW/day, the treatment group (P1), (P2 ), and (P3) only given Solanum betaceum extract at day 1-3 with sequential doses of 100 mg/ kg BW /day, 200 mg/ kg BW /day, and 400 mg/ kg BW /day, then 4-35 days were given Solanum extract betaceum and after 1 hour given lead acetate. The results of this study indicate that there were significant differences between groups with p <0.05. Based on the mean value, administration of Solanum betaceum extract at a dose of 100 mg/ kg BW /day showed the most optimal increase in Leydig cells number.Keywords: lead; antioxidant; Leydig cells; Solanum betaceumABSTRAKPaparan timbal sebagai polusi lingkungan dapat menimbulkan masalah kesehatan reproduksi pria. Pemberian antioksidan alami menjadi solusi alternatif untuk melindungi fungsi reproduksi pria. Penelitian ini dilakukan untuk menemukan potensi ekstrak Solanum betaceum sebagai agen protektif sel Leydig pada mencit yang dipapar timbal asetat. Pelaksanaan seluruh proses penelitian dengan unit replikasi berjumlah 40 ekor mencit jantan. Mencit dibagi dalam lima kelompok dengan setiap perlakuan diberikan secara oral. Kelompok kontrol negatif (K0) hanya mendapatkan aquadest selama 35 hari, kelompok kontrol positif (K1) mendapatkan aquadest hari 1-3, hari 4-35 mendapatkan timbal asetat dengan dosis 0.075 mg/kgBB/hari, kelompok perlakuan (P1), (P2), dan (P3) hanya diberikan ekstrak Solanum betaceum pada hari 1-3 dengan dosis secara berurutan 100 mg/kgBB/hari, 200 mg/kgBB/hari, dan 400 mg/kgBB/hari, kemudian hari 4-35 diberikan ekstrak Solanum betaceum dan setelah 1 jam diberikan timbal asetat. Hasil penelitian ini menunjukkan bahwa terdapat perbedaan yang signifikan antar kelompok dengan nilai p<0.05. Berdasarkan nilai rerata, pemberian ekstrak Solanum betaceum dengan dosis 100 mg/kgBB/hari menunjukkan peningkatan jumlah sel Leydig yang paling optimal.Kata kunci: timbal; antioksidan; sel Leydig; Solanum betaceum
