SUMMARY
En el trabajo se emplea la metodología de cálculo con el MODESLAB a la modelación de la actividad anticoagulante de diferentes fármacos. Para esto se realizó el cálculo de los momentos espectrales de la matriz de adyacencia entre aristas del grafo molecular con hidrógenos suprimidos, ponderada en la diagonal principal con diferentes parámetros a 985 compuestos entre activos e inactivos. Los descriptores calculados fueron usados en una serie de entrenamiento y otra de predicción para obtener y evaluar el modelo respectivamente. Con la serie de entrenamiento se desarrolló una función discriminante para la actividad anticoagulante obteniéndose una buena clasificación total de 92.29 %. El modelo fue validado mediante el uso de una serie de predicción externa con un total de 146 compuestos, obteniéndose una buena clasificación total de un 95.89 %. Se determinó la contribución de enlaces a la actividad (análisis subestructural), De esta forma se obtuvieron las zonas de iso-contribución positiva o farmacóforas, y las zonas de iso-contribución negativa, que pueden actuar como grupos transporte para las moléculas analizadas, lo cual nos da una idea de las regiones que pueden interactuar con un receptor determinado, así como aquellas que facilitan la llegada del fármaco al sitio de acción.Palabras clave: QSAR, anticoagulante, iso-contribución, predicción AbstractIn the work the calculation methodology is used with the MODESLAB to the modeling of the anticoagulant activity of different drugs.
