Abstract

ABSTRAK Penelitian ini diawali dengan menumbuhkan dari daun salam yang telah disterilkan menggunakan etanol 70% dan natrium hipoklorit. Skrining antibakteri dilakukan dengan menggunakan uji antagonis sebagai screening pertama melawan Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis, Salmonella thyposa. Penelitian ini menghasilkan 2 isolat fungi. Fungi endofit aktif difermentasi dan ekstraksi dengan etil asetat (EtOAc) dan metanol. Aktivitas antibakteri dari ekstrak EtOAc dan metanol diuji aktivitasnya menggunakan metode difusi disk. Satu isolat fungi endofit (XP1) terhadap lima dari bakteri yang diuji adalah S. aureus, P. Aeruginosa, E. Coli, S. thyposa, dan B.subtilis, memiliki daya hambat terbesar pada bakteri uji S. thyposa dengan diameter daya hambat sebesar 14,00 mm. Sedangkan pada isolat fungi endofit (XP2) terhadap lima dari bakteri yang diuji adalah S. aureus, P. Aeruginosa, E. Coli, S. thyposa, dan B.subtilis, memiliki daya hambat terbesar pada bakteri uji S. thyposa dengan diameter daya hambat sebesar 22,00 mm. Berdasarkan analisis filogenetik isolat XP 1 memiliki kemiripan dengan Penicillium sp. (homology 87%) sedangkan isolat XP2 memiliki kemiripan dengan Colletotrichium gloeosporioides (homology 98%).

ABSTRACT This research was begun by growing from bay leaves that had been sterilized using 70% ethanol and sodium hypochlorite. Antibacterial screening was carried out using antagonistic tests as the first screening against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis, Salmonella thyposa. This study produced 2 fungi isolates. Active endophytic fungi are fermented and extracted with ethyl acetate (EtOAc) and methanol. Activity. The antibacterial activity of EtOAc and methanol was tested for its activity using the disk diffusion method. One isolate of endophytic fungi XP1 against five of the bacteria tested was S. aureus, P.aeruginosa, E.coli, S. thyposa, and B.subtilis, which had the greatest inhibition on S. thyposa test bacteria with inhibitory diameter of 14.00 mm. Whereas the XP2 endophytic fungi isolates against five of the tested bacteria were S. ureus, P.aeruginosa, E.coli, S.thyposa, and B.subtilis, which had the greatest inhibition on S. thyposa test bacteria with inhibition diameter of 22.00 mm. Based on phylogenetic analysis isolate XP 1 has similarities with Penicillium sp. (homology 87%) while isolate XP2 Colletotrichium gloeosporioides (homology 98%).
